Overview of Centrifugal Spray Dryer:

The centrifugal spray dryer is a core equipment in modern industry that efficiently converts liquid materials into high-quality powders. Its working principle is to instantly disperse and stretch the liquid raw materials through a high-speed rotating centrifugal atomizer, forming a fine mist net. At the same time, hot air is precisely introduced into the drying tower and comes into efficient co-current or counter-current contact with the mist droplets, causing the moisture to evaporate rapidly within a few seconds, and finally obtaining a uniform and dry powder product.

The essence of this process lies in "instantaneous drying". The extremely high heat and mass transfer efficiency enables even heat-sensitive substances (such as proteins, probiotics, or natural extracts) to be dehydrated at a relatively low temperature, thus maximizing the retention of their active ingredients and natural properties. By precisely controlling the atomizer speed, inlet air temperature, and air flow pattern, operators can flexibly control the particle size, moisture content, solubility, and fluidity of the finished powder to meet the strict requirements of many fields, from food milk powder, pharmaceutical raw materials to high-end ceramic powders and nanomaterials.

The centrifugal spray dryer is an extremely important and efficient production equipment in the western medicine industry (especially in the fields of bulk drugs and new formulations). It instantly converts liquid materials into dry powders. Its core principle is to spin the feed liquid into micron-sized mist droplets through a high-speed centrifugal atomizer, which come into co-current or counter-current contact with hot air and complete evaporation and drying within a few seconds, thus obtaining powders with specific particle size and fluidity.

Application of Spray Dryer in Pharmaceutical Production Process:

  1. Active Pharmaceutical Ingredient (API) Production

  • Drying of Heat-Sensitive APIs: Many antibiotics (such as penicillin and streptomycin), vitamins, enzyme preparations, and protein-based drugs are sensitive to temperature. The "instantaneous high temperature, low temperature of the material" characteristic of centrifugal spray drying (the outlet temperature can be precisely controlled, and the actual heating time of the material is extremely short) can minimize the degradation of active ingredients and maintain the drug efficacy.

  • Improving Solubility and Bioavailability: Amorphous drug particles can be prepared by spray drying. Compared with the stable crystalline state, amorphous drugs have higher solubility and dissolution rate, which is crucial for solving the absorption problem of poorly water-soluble drugs (improving bioavailability).

  • Preparation of APIs with specific particle size distribution: By adjusting parameters such as the atomizer rotation speed, feed rate, air temperature, and air volume, the particle size, density, and fluidity of the finished powder can be precisely controlled to meet the direct requirements of downstream formulation processes (such as direct compression and capsule filling), eliminating subsequent processes such as pulverization and mixing.

  • New drug delivery systems and pre - treatment of formulations

    • Microencapsulation and coating: Add drugs and coating materials (such as polymers and lipids) to the feed liquid simultaneously, and form microspheres or microcapsules in one step through spray drying. This can be used for taste masking (such as bitter drugs), enteric/sustained - release control, improving stability, or targeted drug delivery. For example: ibuprofen microcapsules, probiotic embedding.

    • Preparation of solid dispersions: Highly disperse the drug in a hydrophilic carrier (such as PVP, copovidone, HPMC, etc.) and spray - dry them together to form a solid dispersion with molecular - level or ultra - fine particle dispersion. This is one of the most effective strategies to improve the dissolution rate of poorly soluble drugs.

    • Key process for inhalation formulations (DPI): Spray drying is the gold - standard process for preparing inhalable dry powders (such as drugs for asthma and COPD). It can precisely produce spherical particles with an aerodynamic diameter in the range of 1 - 5 microns and excellent fluidity, ensuring that the drug can be effectively deposited in the deep lungs.

    • Preparation of co - processed excipients: Dissolve or suspend two or more excipients (such as lactose and microcrystalline cellulose) together and then spray - dry them to prepare multifunctional composite excipients with better fluidity, compressibility, and stability, which can be directly used in tablet or capsule production.

  • Drying of traditional Chinese medicine and natural drug extracts

      • Although belonging to the category of natural drugs, they are widely used in modern pharmacy. The extract can be dried into powder in one step, which is convenient for making granules, tablets, or capsules and better retains volatile components.

Technical advantages of spray dryers in the pharmaceutical production field:

      1. Extremely fast drying speed: The material contacts with hot air with a huge surface area, and the evaporation of moisture is completed within seconds, which is suitable for continuous and large-scale production.

      2. Excellent product quality: The obtained powder is usually spherical, with good fluidity, uniform particle size, good solubility, and stable chemical properties.

      3. Strong process controllability: By adjusting numerous parameters (inlet air temperature, outlet air temperature, atomizer rotation speed, air volume, properties of the feed liquid, etc.), the physical properties (particle size, density, moisture, morphology) of the product can be finely regulated.

      4. Simplified production process: It realizes the transformation from “liquid to powder” in one step, eliminating multiple unit operations such as evaporation, crystallization, filtration, and pulverization, reducing the risk of contamination and improving production efficiency.

      5. Suitable for aseptic production: The equipment can be designed as a fully enclosed system and equipped with an aseptic filter and an in-place cleaning/sterilization (CIP/SIP) system to meet the production requirements of aseptic bulk drugs or aseptic preparations.
