Core purpose:
Specifically used for sample separation, purification and enrichment before the morphological analysis of alkylmercury such as methylmercury and ethylmercury in samples of environment, food, biology, etc.
1. Morphological separation:
Different mercury morphologies must be separated during alkylmercury analysis. Distillation method is an effective separation means recommended by domestic and international standard methods at present. The distillation device selectively distills alkylmercury such as methylmercury in the sample in the form of chloride through precise temperature control, while inorganic mercury remains in the residual liquid, thus achieving efficient separation.
2. Sample purification and enrichment:
The matrix of samples to be tested (such as sediment, fish meat, tissue, water body) is complex, containing a large amount of protein, fat, pigment and other organic/inorganic impurities, which will seriously interfere with subsequent analytical instruments (such as HPLC-ICP-MS/AFS).
The distillation process can effectively get rid of the interference of complex matrix and obtain a pure alkylmercury fraction.
By controlling the volume of the receiving liquid, the concentration and enrichment of the target substance can be achieved, improving the sensitivity and detection limit of the analytical method.
3. Preparation for subsequent detection:
The pure fraction collected by distillation can be directly or after derivatization, enter instruments such as high performance liquid chromatography - atomic fluorescence spectrometry (HPLC-AFS), high performance liquid chromatography - inductively coupled plasma mass spectrometry (HPLC-ICP-MS) for qualitative and quantitative analysis.
Choosing PFA as the material for the distillation flask is the key to complete the above tasks, and its advantages are incomparable to those of glass or other plastic materials:
1. Ultra-high chemical inertness
· Advantage: It can withstand all strong acids required in the whole distillation process, including concentrated acids, such as hydrobromic acid, hydrochloric acid, sulfuric acid, nitric acid. It ensures that there are no dissolved substances in the high-temperature acidic environment and no impurities are introduced to interfere with the determination.
· Comparison: Ordinary glass will precipitate silicate and various metal ions under such conditions, seriously polluting the sample.
2. Low background value of metal ions
· Advantage: The content of metal ions in PFA is very low, which is crucial for the morphological analysis of trace mercury, ensuring the ultra-low background and ultra-high accuracy of the analysis results and effectively preventing false positives or excessive background values.
· Comparison: Even borosilicate glass contains a detectable background value of mercury.
3. Excellent anti-adsorption properties
· Advantage: The surface of PFA is smooth and dense, with very low adsorption of target analytes such as methylmercury, which can maximize the recovery rate of fractions and the reliability of results. There is no residue after cleaning, avoiding cross-contamination.
· Comparison: The glass surface has a strong adsorption effect on mercury compounds, resulting in a low and unstable recovery rate.
4. Good thermal stability and transparency
· Advantage: PFA material can withstand high and low temperatures (-200 to 250℃) under normal pressure, meeting the temperature requirements for the distillation of methylmercury and alkylmercury (usually between 140-180℃). At the same time, PFA material has a certain degree of transparency, which is convenient for experimenters to observe the boiling, reflux and distillation processes of the solution in the bottle.
5. Not easy to break
· Advantage: Compared with glass, PFA has better toughness and impact resistance, is not easy to break due to collisions or thermal stress, has a longer service life and higher safety.
