Instrument Features:

Air Circuit Schematic:

1. The test device mainly consists of an aerosol generation device, aerosol chamber, sampler, and vacuum source.

1.1 The core components of the aerosol generation device are an imported UK aerosol generator and Japanese imported pressure control unit. The pressure control unit regulates the gas pressure applied to the aerosol generator to produce aerosol of suitable concentration and particle size. Gas pressure control to 0.1kPa, adjustable in real-time during experiments.

1.2 Considering that peristaltic pump dosing at 0.01ML/MIN is intermittent, a reservoir-type aerosol generator is chosen. Liquid medicine can be directly injected into the generator and adjusted via the spray nozzle.

1.3 Positive pressure source is provided by an oil-free air compressor, filtered through an air filter, and connected to the pressure control unit. Pressure adjustable from 5kPa~500kPa.

2. The aerosol chamber is manufactured according to Figure B.2, with two sampler interfaces and reserved filter interfaces.

3. The samplers are two Anderson samplers. The downstream of the sampler is controlled by a flow switch with adjustable flow rate, range 0~50L/min adjustable. Gas drying is required between the sampler and flow meter.

4. The vacuum source is provided by two vacuum pumps, each capable of providing negative pressure gas flow ≥30L/MIN.

Program Functions:

1. Open setting for each experimental step: Configure parameters for all steps in the experiment on a unified timeline, such as generator working time, sampler working time, etc., all with open start and stop times. Allows flexible and intuitive combination of experimental processes.

2. Negative pressure inlet pressure protection function, real-time monitoring of positive pressure inlet pressure to ensure gas demand for the aerosol generator is met during experiments, guaranteeing qualified aerosol production. Otherwise, alarm and stop experiment.

3. Negative pressure source protection function, real-time monitoring of negative pressure and flow during experiments to ensure standard test conditions are met. Otherwise, alarm and stop experiment.

4. Positive pressure gas automatic control function. Just input the pressure required for generator operation, the system's internal algorithm automatically performs feedback pressure adjustment to quickly stabilize at the desired pressure.

Experimental Procedure:

1. Turn on the air compressor and vacuum pump power and valves for inflation.

2. Connect all piping sections.

3. Connect the Anderson samplers.

4. Add test bacterial solution to the sprayer.

5. Set test parameters in the parameter setting interface according to the experimental times in standard B.4.

6. Follow the steps in B.4: perform positive control, sample challenge, and negative control experiments sequentially.

7. Culture the agar plates.

8. Clean the inside of the equipment and piping for the next experiment.
