  Bacterial Vaginosis Quintuplet Diagnostic Kit (Chemical Reaction)

Intended Use

Bacterial Vaginosis Diagnostic Kit is adopted to test catalytic activity of N-Acetyl-β-glucosaminidase, Sialidase, Leukocyte Esterase and β-Glucuronidase, Perhydrol (H₂O₂) concentration and pH value in vaginal secretion, it’s disposable and used for screening and auxiliary diagnosing vaginitis.

Assay Principle

In the chemical inspection of vaginal secretion, chromogenic reaction will take place if the specific metabolite takes reaction with the specific substrate. The chromogenic reaction of each separated testing item determines the existing of BV, Monastery Vaginitis, Colpitis Mycotica, bacillus acidi lactici, and denotes pH value.

N-Acetyl-β-D-glucosaminidase (NAG) Activity Detection: NAG is the enzyme secreted by viruses like Monilia, Trichomonas vaginalis and Mycoplasma hominis.  Healthy vagina secretion doesn’t contain NAG. So NAG chromogenic reaction test will tell whether the vagina has been infected with those viruses. 

The Sialidase (SNA) Activity Detection: SNA is the specific enzyme secreted by pathogenic bacterium like gardnerella vaginalis, mobiluncus etc. The positive result denotes bacterial viginosis. 

Leukocyte Esterase (LE) Activity Detection:  Leukocyte Esterase is the carboxylesterase released by polymorphonuclear leucocytes. When inflammation takes place, polymorphonuclear leucocytes will gather at the focus and release more leukocyte esterase. So too much polymorphonuclear leucocytes denotes bacterial and fungal vaginal infection. The LE activity reflects vaginal cleanliness and assists the diagnosis of vaginitis.

Perhydrol (H2O2) Concentration Detection: Perhydrol is the specific product of vaginal bacillus acidi lactici. Its concentration indicates whether the beneficial bacterium is normal in vagina. The negative result denotes that the bacillus acidi lactici which maintains vaginal microecological balance is normal while the positive means that the microecology is imbalance.

pH Value: pH value is auxiliary index for clinical diagnosis of vaginal infection, it doesn’t have specificity. The vagina pH value of a healthy woman is 3.8 - 4.5. The negative denotes that the bacillus acidi lactici which maintains vaginal microecological balance is normal while the positive means that the balance was broken and the microecology is imbalance.

Storage Condition and Period of Validity

Storage Condition: stored in the 2℃~8℃ circumstance without non corrosive substances. Never freeze it.

Interpretation of Results

1. NAG (β-N-acetyl-glucosaminidase):NAG without color denotes negative, indicating normality. NAG displaying yellow denotes positive, the mucin barrier on surface of vaginal mucosa was damaged, vaginal mucosa is susceptible to infection or infected by Monilia Albanian,Trichomonas Vaginalis, Mycoplasma Hominis etc.

2. SNA (Sialidase):SNA without color denotes negative, indicating normality. SNA displaying blue or green denotes positive, indicating bacterial vaginosis.

3. LE (Leukocyte Esterase): LE without color denotes negative, indicating normality. LE displaying blue or green denotes positive, there is inflammation.

4. H₂O₂(Perhydrol): H₂O₂ indicating fuchsia denotes negative, indicating normality. H₂O₂ without color denotes positive, indicating that flora is in imbalance.

5. pH value: Interpret it according to Standard Color Card.
