Based on the popular LS-50B, the LS 45 spectrometer has been developed in conjunction with a host of accessories and software to address a wide range of applications. Whether your application requires fluorescence, phosphorescence, chemi - or bioluminescence, the LS 45 Fluorescence spectrometer is ready for the challenge.
For the routine user, this fixed slit instrument provides a robust, reliable system for the rigors of day-to-day use. The LS 45 is an instrument with all of the heritage, sensitivity and reliability you've come to expect, but with surprising flexibility at a very reasonable price.

This monochromator based instrument uses a high energy pulsed Xenon source for excitation. For more than 20 years PerkinElmer has relied on pulsed Xenon sources to minimize photo bleaching of samples and provide a long-lived excitation source.
FL WinLab™ software, designed for the demanding needs of modern laboratories, operates in the Microsoft® Windows® environment and controls the LS 45. The software combines the user-friendly appeal of a Windows based software package with application specific knowledge PerkinElmer has developed through decades of experience. The result is a powerful system that can be used routinely for quality fluorescence analyses.
Specific modes of instrument operation such as Scan, Time Drive, and Ratio Data Collection can be easily accessed in the Application Menu. The excitation and emission monochromators can be scanned independently or synchronously. In addition, the PRESCAN mode is an ideal method development tool for location of peak excitation and emission maxima. FL WinLab includes a validation protocol that automatically checks the instrument performance to ensure that it is operating within specification. FL WinLab, when combined with the LS 45, provides a wealth of tools for data collection and analysis such as:
- Fluorescence, Phosphorescence and Bio- and Chemiluminescence measurement modes
- Spectral scanning, including excitation, emission, constant wavelength synchronous and constant energy synchronous
- 3D Excitation/Emission (EEM) and 3D synchronous scanning
- 3D kinetic scanning
- Single- and multiple-wavelength kinetics
- Simple quantitation by curve fitting with a number of fit algorithms
- Intracellular ion analyses
Accessories add sampling flexibility
The LS 45 includes a single cell thermostatted sample holder. The cell holder accommodates 1 cm pathlength cells as well as semi-micro cuvettes. Semi-micro cuvettes are particularly useful where the sample is precious or the volume is limited such as cell cultures and DNA samples. A variety of accessories are available for use with the LS 45. For example, our automated Polarizer consists of 2 filter wheels; each wheel containing a horizontal and vertical polarizing element. Polarizer positions are software controlled and can be manually set or automatically controlled for polarization, anisotropy or G-factor. Additional accessories include the following: Application figure window. Single cell thermostatted sample holder is included with the LS 45.
Front Surface accessory
The Front Surface accessory is ideal for analyzing films, papers, powders and other flat samples. The sample may be placed in the accessory directly, or held in the powder holder which has a synthetic fused silica window. Ultra small volumes or viscous samples such as oils can be sandwiched between two windows and placed in the holder. In addition, opaque and turbid samples can be measured.
Remote Fiber Optic accessory
The Remote Fiber Optic accessory allows you to take the measurement to the sample rather than taking the sample to the instrument. It is ideal for remote, non-destructive testing of fluorescent papers and fabrics, and remote sampling of hazardous materials such as radioactive sources.
LC Flow Cell
The LC Flow Cell is ideal for high sensitivity liquid chromatography, and is a good way to expand the utility of the LS 45. This solution provides laboratories with an alter-native to a dedicated LC fluorescence detector. The twin monochromators of the LS 45 enable accurate wavelength selection for maximum selectivity and sensitivity.
AS93plus Autosampler accessory
The high throughput AS93plus Autosampler, when coupled to the LS 45, can measure up to 200 samples in a single rack. This provides a high sensitivity alternative to microplate reading. The Autosampler has a built-in peristaltic pump that is controlled by our FL WinLab software.
